The Opportunity Isle Soul is a random drop from opening the Angler's Pouch, a reward for participating in the island's fishing contest. The winner on the island will receive three of these, while all other players receive either two or one, depending on their placement.

A good trick to fish a little faster is to not use Net Fishing and keep your stacks. This allows you to pull fish up slightly faster. It's worth it to get up to 5 stacks of the buff before the contest starts as well if you're trying for the Island Soul.
